Activity Definition: One unit of enzyme activity is defined as the amount of enzyme that causes an increase in absorbance of 0.001 per minute at 260 nm at 25°C when catalyzing a high molecular weight DNA solution.

Appearance: White or off-white lyophilized powder. It is a double-strand-specific endonuclease that degrades double-stranded DNA into oligodeoxyribonucleotides with 5′-phosphate and 3′-OH termini. In the presence of monovalent manganese ions, it can also degrade single-stranded DNA. Soluble in water, slightly soluble in 30% ethanol and acetone. Enzyme reaction: Deoxyribonucleic acid → dinucleotide-5′-phosphate + oligonucleotide-5′-phosphate. Optimal pH 7.8. Extinction coefficient (E1%280) 11.1. Activators include divalent metal ions, with maximum activation achieved by magnesium (Mg²⁺) + calcium (Ca²⁺). Inhibitors include EDTA and sodium dodecyl sulfate. May be supplemented with 5 mmol/L calcium (Ca²⁺).

Usage: Biochemical research. Used in molecular biology research. Degrades various enzymes of DNA. These enzymes hydrolyze phosphodiester bonds in the sugar-phosphate backbone. Such enzymes are divided into two types: exonucleases and endonucleases. There are many types of DNases, some being single-strand-specific, some double-strand-specific, or sequence-specific (e.g., restriction endonucleases). Restriction endonucleases have the ability to recognize specific base sequences, a property utilized in genetic engineering techniques to construct artificial clones.
